Clinical presentation of MEN 2A in index vs. non-index patients - Endocrine
Source : https://link.springer.com/article/10.1007/s12020-023-03459-8
Purpose Differences in syndromic manifestations of multiple endocrine neoplasia 2 A (MEN2A) between index and non-index patients are ill-defined. Methods Cross-sectional analysis of 602 REarranged during Transfection (RET) carriers (156 index...
Conclusion: These findings imply that differences between index and non-index patients impact the first syndromic manifestation without extending to subsequent syndromic manifestations. Because they exhibited similar age and tumor characteristics for the secondary and tertiary manifestations of MEN2A, screening for these syndromic components...
Challenges in diagnosis and biomarker testing for RET-altered lung and thyroid cancer care: an international mixed-method study - BMC Medical Education
Source : https://bmcmededuc.biomedcentral.com/articles/10.1186/s12909-023-04396-w
Background The introduction of new targeted therapies for RET-altered lung and thyroid cancers (LC/TC) has impacted pathologists' practice by making genomic testing more relevant. Variations in health systems and treatment...
Conclusions: This study identified areas where pathologists need additional continuing professional development opportunities to enhance their competencies and better support delivery of care to patients with RET-altered lung or thyroid tumours. Addressing identified gaps and improving competencies of pathologists in this field should be...
Loss of Tumour Suppressor TMEM127 Drives RET-mediated Transformation Through Disrupted Membrane Dynamics
Source : https://www.biorxiv.org/content/10.1101/2023.06.28.546955v1
Internalization from the cell membrane and endosomal trafficking of receptor tyrosine kinases (RTK) are important regulators of signaling in normal cells that can frequently be disrupted in cancer. The adrenal...
Conclusions/Relevance: Together, our data identify TMEM127 as an important determinant of membrane organization, including membrane protein diffusability and protein complex assembly, and provide a novel paradigm for oncogenesis in PCC where altered membrane dynamics promotes cell surface accumulation and constitutive activity of growth...
Rapid and Cost-Efficient Detection of RET Rearrangements in a Large Consecutive Series of Lung Carcinomas - PubMed
Source : https://pubmed.ncbi.nlm.nih.gov/37445709/
RET -kinase-activating gene rearrangements occur in approximately 1-2% of non-small-cell lung carcinomas (NSCLCs). Their reliable detection requires next-generation sequencing (NGS), while conventional methods, such as immunohistochemistry (IHC), fluorescence in situ...
Conclusions/Relevance: The results of our study provide a viable alternative for RET testing for those facilities that do not have access to NGS due to cost or technical limitations. The proposed pipeline requires validation in independent laboratories.
Review article: new treatments for advanced differentiated thyroid cancers and potential mechanisms of drug resistance
Source : https://www.frontiersin.org/articles/10.3389/fendo.2023.1176731/full
The treatment of advanced, radioiodine refractory, differentiated thyroid cancers (RR-DTCs) has undergone major advancements in the last decade, causing a paradigm shift in the management and prognosis of these patients.
Conclusions/Relevance: In this review, we will discuss currently available drugs for advanced RR-DTCs, potential mechanisms of drug resistance and future therapeutic avenues.
