A narrative review about CDK4/6 inhibitors in the setting of drug resistance: updates on biomarkers and therapeutic strategies in breast cancer

Conclusions: This review clarified the current knowledge about mechanisms, the biomarkers to overcome the drug resistance of CDK4/6 inhibitors, and the latest clinical progresses about CDK4/6 inhibitors. Possible approaches to overcome CDK4/6 inhibitors resistance were further discussed. For example, using another CDK4/6 inhibitor, PI3K...

Clinical Management of Potential Toxicity of Abemaciclib and Approaches to Ensure Treatment Continuation

Clinical Management of Potential Toxicity of Abemaciclib and Approaches to Ensure Treatment Continuation

Source : https://journal.waocp.org/article_90662.html

Introduction: The association between abemaciclib dose reduction and treatment adherence is not clear. In this study, we examined real-world data of Japanese patients with advanced breast cancer (ABC) to determine...

Conclusions: In this study, the 100 and 200 mg/day groups had a longer TTF than the 300 mg/day group, and dose reduction was identified as an important factor in achieving longer TTF.

CDK4/6 Inhibitors: Existing and Emerging Differences - PubMed

CDK4/6 Inhibitors: Existing and Emerging Differences - PubMed

Source : https://pubmed.ncbi.nlm.nih.gov/37369022/

The cyclin-dependent kinase (CDK)4/6 inhibitors palbociclib, ribociclib, and abemaciclib are standard-of-care therapy for hormone receptor-positive advanced/metastatic breast cancer, based on randomized trials showing improved progression-free survival for all 3 drugs,...

Conclusions/Relevance: We focus particularly on how emerging findings may help us to understand similarities and differences between the available CDK4/6 inhibitors. Even at late-stage clinical development, there remains much to learn about how agents in this class exert their varying effects.

Real-world data analysis of perioperative chemotherapy patterns, G-CSF use, and FN status in patients with early breast cancer - Breast Cancer Research and Treatment

Real-world data analysis of perioperative chemotherapy patterns, G-CSF use, and FN status in patients with early breast cancer - Breast Cancer Research and Treatment

Source : https://link.springer.com/article/10.1007/s10549-023-07015-w

Purpose This study aimed to describe perioperative chemotherapy patterns, granulocyte colony-stimulating factor (G-CSF) use, and febrile neutropenia (FN) status in patients with early breast cancer (EBC) using real-world data in...

Conclusion: Despite the increasing use of escalated regimens in the last 5–6 years, FNH continuously declined, and the odds of FNH were lower among patients treated with pegfilgrastim PP. These results may suggest the contribution of PP in part to suppressing FNH levels over the last 5–6 years.

Disproportionality Analysis of Abemaciclib in the FDA Adverse Event Reporting System: A Real-World Post-Marketing Pharmacovigilance Assessment - Drug Safety

Disproportionality Analysis of Abemaciclib in the FDA Adverse Event Reporting System: A Real-World Post-Marketing Pharmacovigilance Assessment - Drug Safety

Source : https://link.springer.com/article/10.1007/s40264-023-01334-z

Background and Objective Abemaciclib, a cyclin-dependent kinase 4 and 6 inhibitor, demonstrated efficacy in women with hormone receptor-positive, human epidermal growth factor receptor 2-negative advanced breast cancer. Because of the...

Conclusions: The discovery of disproportionality signals could potentially prompt improved awareness of toxicities for abemaciclib, and the results of time to onset, serious and non-serious reports, and clinical priority analyses provided some supporting evidence for clinicians to manage adverse events.

I find this agent a bit too toxic at full dose so it is probably not a bias but the truth
